95% sure. Obviously we won't know for certain until we see the movie.
But several spoiler reports from different people and things people have posted on imdb suggest that the two blondes are killed in Stab 10 (Not sure on the number but I know it's in a Stab film which suggest they're showing how the Stab films kept going to poke fun at the endless remakes and sequels in Hollywood).

Anyway once it's revealed to be the Stab film, it cuts to one of Scream 4's main characters watching the movie on DVD and then the actual opening kill occurs. The killings begin during "Remembrance Week" at Woodsboro High when Sidney is launching her new book.

I'm pretty sure I heard something along the lines of a Stab 7. The Stab films are kept going by Gale who, since the events of Scream 3 had written some Stab novels that have been adapted onto film.

I don't think Courtney and David's split will overshadow the movie, I think they'll use it to get the movie more attention. Every article / tv spot that talks about their split will contain the added line; "Who recently finished filming Scream 4", "who will star in the fourth installment of the Scream franchise released April 2011" etc.

And I've been on the imdb boards and stupidly a lot of people are screaming "OMG, Gale's the killer!" because of the shot that shows her taking a mask off. As if they would actually spoil the killer's identity in the promo.

Also, have any of you heard the rumours about Kevin Williamson cutting ties with the movie and a possible 5 and 6. Apparently they made some major changes to his script during shooting and he wasn't happy that they didn't consult him.

Yeah I think if 4 is a success then there should be no reason not to do a 5th and 6th one. The cast looks okay. A bit "young" though with Emma Roberts in and that. I hope they are decent at acting in a horror film.

Since the trailer was leaked a friend and I have been speculating a lot of things about this movie including some deaths and already who we think the killer might be lol. When I say 'A lot' I mean a lot so don't read if you want to remain completely surprised and it turns out one of these theories is correct.

Spoiler:

Theory 1) Gale dies! I think someone reported Courtney Cox finished filming early and this could be because of her busy schedule on Cougar Town, however, the released pics and the trailer seem to show a lot of that scene where she says "Do it if you have the guts!" to the killer. It makes me think she might not be in the film much at all and that this scene could very well be Gale's death scene. Hope not though

Theory 2) Sidney or Jill (Emma Roberts) die. Sidney because it could mark the end of her reign as the Scream queen and have over the baton to Jill seein as Neve Campbell was apprehensive about doing Screams 2 and 3 as well. Or, perhaps, if Jill is just a carbon copy of Sidney (being virginal in this film) and with the new rule that virgins can now die, maybe she'll bite the dust because she simply isn't Sidney.

Theory 3) Sidney or Jill is the killer! Sidney because Wes Craven claims Sidney's bene living in peace for the last decade and now has to deal with more killings. Perhaps this is a red herring and she's now as insane as everyone who tried to kill her and could relate back to that bathroom scene in the first film when the bitchy school girl said "It ******ed her up royally." So, another way for Sidney to hand over the reigns. Or, seeing as how ******ed up her family is, and because this isn't a straightforward remake of the original Scream, Jill could be the killer.

Theory 4) Trevor (Nico Tortorella) is the killer, or one of the killers. Seeing as Scream 4 will parody more sequels and recent remakes you'll notice Scream 4 is a sort of remake of the first film apart from Sidney, Gale and Dewey being in it. Jill is the 'remake' Sidney, Hayden Panettiere's Kirby is the new Tatum, Rory Culkin's Charlie is the new Randy, and Trevor is the new Billy. Maybe there's another Stu out there helping Trevor re-enact Billy and Stu's murders?

Theory 5) Marley Shelton's character is the killer! Her character is described as "Having known Sidney during high school", and back when it was reported Lake Bell was going to play this character it was suggested she cares about her boss Dewey, "Perhaps a bit too much". If Gale does die (Theory 1) the motive for Shelton's character could be to get her out of the way in order to get to Dewey or something like that. Maybe she even kills Dewey? If she can't have him, no one can?
Also, originally Angelina was going to be the second killer in Scream 3 and her story was that she'd be Roman's girlfriend and knew Sidney from high school. However, it was decided Roman's romance with the actress playing Sidney (his own sister) in the 'Stab' movies would be a bit too weird. Maybe Kevin Williamson's recycled this abandoned idea from Scream 3?

They've already said they're planning to do 3 more bringing it up to Scream 6.

I remember reading it when I was reading about A Nightmare On Elm Street being brought up to 2014 meaning there'd be two more.

And then Scream was mentioned to have a 'new trilogy'.

Just hope Scream 4, Scream 5 and 6 are all good and it doesn't die.

Fun Fact: The original script of Scream 2 said that Hallie was to be one of the murderers, along with Derek (who she was in a secret relationship with) and Mrs. Loomis. Had the script not changed, Hallie would have been the only African American Ghostface.

It also kinda refers back to one of the theories.

I hops Scream 4's as good as 1-3 and that there'll eb another 2 but what what the next 2 be about? Scream 1 obviously mocked/homaged horror movies, Scream 2 parodied sequels, Scream 3 was more about satirizing Hollywood in general and a bit about trilogies and Scream 4 will eb about endless sequels (again) and remakes so where does that leave Scream 5 and 6? Prequels, sidequels (spin-offs) and straight to DVD? God I hope not to any of them lol

I knew that and I'm glad Hallie and Derek weren't the second and third killer. I love Scream 1 and 2 but from what I gather Kevin Williamson is quite flawed. In the commentary for Scream 1 he admits he only killed Principal Himbry because the Weinsteins told him he needed another death scene and he hadn't really thought about why a lot of the teenagers would have left the party.
Also, I hope the killer in Scream 4 is a woman because otherwise Mrs Loomis will remain the only female killer

Well referring to someone's theory about Gale dying. I would be pretty pissed off if that was the case and they'd put her dying words in the teaser trailer. That's pretty lame.

Also, after watching the teaser and hearing the new rule about the killer wanting to film the murders. (Don't know how that's new, they've done that in movies for years), it's making me think that Gale sets that scene up. Not because she's the killer but because she is running out of ideas for her writing and she's maybe not as famous as she used to be so she fakes a Ghostface attack and posts it on the internet without realising that Ghostface is in fact back and hears about whatever actual murder.
Probably not true but it's a possibility. Also, don't think Courtney finishing filming early says anything about ow much of the movie she's in because I heard a rumour that they filmed different versions of the climax/reveal scenes early on during shooting.
